How do you get tickets to JLeno show

Do you need to stand in line? If I know that I will be in LA on
such-n-such date, is there a way I can get tickets. If not, what time
do you need to be in line to guarantee a ticket?

I think his show is near Universal Studios. Am I correct?

Yes, unless you know somebody at NBC who can get passes for you.

I think they mail tickets to out-of-town addresses (check the
website for details). But keep in mind that those "tickets" only
let you wait in line for the show, they don't guarantee you a seat.

The line starts forming early in the morning (often before 8 AM),
or even days before if a 'hot' musical group is scheduled to appear.
